Utilizing Peircean semiotic theory, this study examines the semiotic building blocks of a festival tourist destination, taking the Guangfu Temple Fair in China as its primary focus. An investigation utilizing grounded theory, a qualitative research approach, was conducted on the organizers' planning scheme, conference materials, seven organizer interviews, and forty-five tourist interviews. Festival organizers' festivalscape design is shaped by social values and tourist expectations, incorporating aspects such as safety assurance, cultural experiences, quality personnel service, facilities, creative interactions, food options, trade shows, and the general festival atmosphere. Tourists' comprehension of a festival's appeal, driven by cultural, innovative, social, and emotional experiences along with incidental observations, rests on recognizing cultural diversity, lively events, prominent features, and a celebratory atmosphere. The conceptual model underpinning the semiotic construction of festivals as tourist attractions is based on how organizers produce signs and how tourists interpret those signs. Beyond that, the research increases understanding of tourist attractions and empowers organizers in constructing profitable festival attractions.
Gastric cancer with PD-L1 positivity is currently treated most effectively by the combination of chemotherapy and immunotherapy. Unfortunately, a definitive and optimal course of treatment for elderly or delicate gastric cancer patients has yet to be established. Previous examinations of the subject matter have ascertained that PD-L1 expression, the presence of the Epstein-Barr virus, and high microsatellite instability (MSI-H) are probable prognostic indicators for the effectiveness of immunotherapy in gastric cancer patients. The Cancer Genome Atlas gastric adenocarcinoma data demonstrated a statistically significant increase in PD-L1 expression, tumor mutation burden, and MSI-H frequency in elderly (over 70) gastric cancer patients compared to their younger (under 70) counterparts. The presence of diabetes is linked to a higher likelihood of kidney disease, heart failure, and an increased risk of death. Sodium-glucose cotransporter 2 inhibitors (SGLT2i) effectively impede these adverse outcomes; however, the precise mechanisms are not yet established. We crafted a comprehensive roadmap of metabolic alterations in different organs due to diabetes and the influence of SGLT2i. Following in vivo treatment with or without dapagliflozin, normoglycemic and diabetic mice underwent metabolic labeling with 13C-glucose, metabolomics, and metabolic flux analysis. Results indicated that glycolysis and glucose oxidation were impaired in the kidney, liver, and heart of the diabetic mice. The application of dapagliflozin treatment failed to reverse the glycolytic deficiency.
